Biologic Agents

Targeted biologic therapies offer new avenues of treatment options for patients with advanced/metastatic breast cancer. In particular, HER2-targeted agents play a definitive role in improving survival in patients with metastatic breast cancer. In addition to single-agent activity, these agents show potent synergy with many chemotherapy agents in this setting. Agents targeting the EGFR and VEGF pathways are also under investigation, as are other novel agents targeting distinct cellular growth or survival pathways. Alone or in combination with conventional cytotoxics, these agents promise to add to the number of active treatment options. Current challenges include optimizing patient selection, integrating these new agents into traditional chemotherapy regimens, and determining the most effective combination regimens with the least toxicities.
